The InCELL Hunter U2OS Bcl2L1-Bax Protein Binding Cell Line provides an easy-to-use cell based assay to study small molecule drug candidates that disrupt protein binding. This kit assesses compound-based disruption of Bcl2L1-Bax binding via detection of Bax levels. This product uses EFC technology. The cell line included overexpresses Bcl2L1 target in an untagged form and Bax binding partner fused to ProLabel. When a compound engages the Bcl2L1 target protein, partner binding is disrupted and the destabilized Bax is degraded, leading to a loss of EFC signal upon addition of EA containing detection reagent. The included cell line overexpresses EA-tagged TRHR and a ProLink-tagged early endosome protein. Upon ligand-induced receptor activation, resulting internalization of the TRHR receptor to the early endosome leads to production of EFC signal.
